藥理作用
積雪草為傘形科植物積雪草(Centellaasiatica(L)Urban)的干燥全草或帶根全草,又稱雷公根、馬蹄草、落得打,常年可采,資源豐富,生長(zhǎng)在東南亞、澳大利亞、南非和中非熱帶地區(qū),在很多國(guó)家的醫(yī)藥領(lǐng)域中應(yīng)用已有悠久歷史。積雪草性寒,味苦,具有清熱利濕、解毒消腫之功效,主要用于濕熱黃疸、中暑腹瀉、癰腫瘡毒、跌打損傷等癥。
積雪草苷(Asiaticoside)屬于五環(huán)三萜類(lèi)化合物,具有抗?jié)?、促進(jìn)傷口愈合、抗腫瘤、抗炎、免疫調(diào)節(jié)等多方面的藥理作用。目前主要用于治療硬皮病和皮膚的創(chuàng)傷及燒傷。
作用機(jī)制
積雪草苷可作用于成纖維細(xì)胞的細(xì)胞核,使核分裂相減少,核仁變小或缺失,隨著用藥濃度的增加,細(xì)胞內(nèi)DNA合成減少,細(xì)胞生長(zhǎng)呈抑制狀態(tài),抑制率最高可達(dá)73%,提示積雪草苷的作用機(jī)制是阻斷成纖維細(xì)胞的增殖,從而減少膠原蛋白的合成,阻止瘢痕增生。
應(yīng)用
積雪草苷(Asiaticoside)是藥材積雪草中的主要成分,積雪草苷A與積雪草苷B的混合物具有促進(jìn)傷口愈合、抗皺、抗炎、抗抑郁、促進(jìn)智力等藥理作用。比較積雪草苷促進(jìn)傷口愈合,治療疤痕的藥理作用可以發(fā)現(xiàn),積雪草總皂苷的抗炎、抗抑郁、促進(jìn)智力等藥理作用可能是積雪草苷A與積雪草苷B起了主要作用。
制備方法
稱取500g粉碎積雪草粉末,以2500ml質(zhì)量濃度為75%乙醇水溶液浸泡,在50℃條件下超聲波提取4次,每次1小時(shí),過(guò)濾,將濾液合并減壓濃縮得到浸膏20.5g;用200ml水溶解浸膏,用浸膏水溶液等體積的石油醚萃取3次,下相水溶液再用等體積的乙酸乙酯萃取4次,匯集下相水溶液。用匯集下相水溶液同等體積的正丁醇與乙酸乙酯體積比為1∶1混合溶劑萃取3次,合并上相萃取液,用活性碳脫色萃取液,減壓濃縮脫色后的萃取液得積雪草總苷7.3g;加入10倍量甲醇溶解積雪草總苷,過(guò)濾,濾液濃縮后,再用質(zhì)量濃度為90%乙醇水溶液在室溫條件下重結(jié)晶得到高純度的積雪草苷3.3g,純度為90%,得率0.66%。
