Reaction Conditions:
Stage #1:α-bromoacetophenone with sodium azide in water;acetone at 20; for 2 h;Cooling with ice;
Stage #2: with hydrogenchloride;palladium on activated charcoal;hydrogen in methanol;water at 20; for 22 h;Cooling with ice;
Steps:
10 2-amino-1-phenylethan-1-one(compound 26)
General procedure: Phenacyl bromide (25) (8.9 g, 44.56 mmol)Acetone (89.1 mL)- water (44.5 mL)Sodium azide (3.5 g, 53.48 mmol) was added under ice-cooling,And the mixture was stirred at room temperature for 2 hours.After distilling off acetone,Extraction with diethyl ether,The organic layer was concentrated.The residue was dissolved in methanol (178.2 mL) and concentrated hydrochloric acid (19.7 mL)Palladium-carbon (4.7 g, 4.45 mmol) was added under ice-cooling,Under a hydrogen atmosphere,And the mixture was stirred at room temperature for 22 hours.The mixed solution was filtered and concentrated,The residue was recrystallized from diethyl ether-methanol to give compound 26 as a white solid (5.8 g, 76%).
