600 parts of formic acid (85%), 225 parts of 2-methyl-5-nitroimidazole and 250 parts of dimethyl sulfate are heated at reflux (80° C) for 4 hours in a stirred vessel, after which the formic acid is distilled off in vacuo. The remaining residue was dissolved in 400 parts of water, neutralized with ammonia water to pH 1.8, and cooled to 0-5°C. Unreacted 2-methyl-5-nitroimidazole was deposited and centrifuged. Then the pH of the mixture was adjusted to 10 with ammonia solution, and 1,2-dimethyl-5-nitroimidazole was separated at 10°C to obtain 75 parts of 2-methyl-5-nitroimidazole and 145 parts of 1,2-dimethyl-5-nitroimidazole. The reaction yield of 2-methyl-5-nitroimidazole was 87% of the theoretical yield.
