蛋白結構(Molecular Characterization)

This protein carries an Avi tag (Avitag?) at the C-terminus, followed by a polyhistidine tag. This protein undergoes autocatalytic cleavage to release the pro-peptide and mature chain. The protein has a calculated MW of 13.9 kDa and 61.1 kDa. The protein migrates as 16 kDa and 66 kDa when calibrated against Star Ribbon Pre-stained Protein Marker under reducing (R) condition (SDS-PAGE) due to glycosylation.

前蛋白轉化酶枯草桿菌蛋白酶/kexin 9型(PCSK9)也稱為NARC1(神經(jīng)凋亡調節(jié)轉化酶),是一種新鑒定的屬于肽酶S8亞家族的枯草蛋白酶。小鼠PCSK9被合成為可溶性酶原,并在內質網(wǎng)中經(jīng)歷自催化分子內加工,導致其前肽被切割,該前肽仍與分泌的活性酶相關,具有廣泛的堿性pH最佳值。這種蛋白質在膽固醇穩(wěn)態(tài)中起著重要的調節(jié)作用。PCSK9與低密度脂蛋白受體(LDLR)的表皮生長因子樣重復序列A(EGF-A)結構域結合,誘導LDLR降解。PCSK9也可能在皮質神經(jīng)元的分化中發(fā)揮作用。該基因的突變與一種罕見的常染色體顯性家族性高膽固醇血癥(HCHOLA3)有關。
