IPTG, Dioxane-Free (isopropyl-β-thiogalactoside) is an analogue of allolactose that binds specifically to the repressor protein of the lac operon and induces expression of β-galactosidase in Escherichia coli. The compound is widely used with X-Gal plates to screen for blue and white colonies. The lac operon system and IPTG have been used in multiple cloning experiments and cellular systems to study gene activity.IPTG 常用于需要誘導(dǎo) β-半乳糖苷酶活性的克隆實(shí)驗(yàn)。它常與 X-Gal 或 Bluo-Gal 結(jié)合使用，用于重組細(xì)菌菌落的藍(lán)白篩選，這些菌落可以誘導(dǎo) lac 操縱子在大腸桿菌中的表達(dá)。IPTG 與 lacI 阻遏蛋白結(jié)合并改變其構(gòu)象而發(fā)揮作用，防止 β-半乳糖苷酶編碼基因 lacZ 的抑制。
