Into a 150 mL sealed tube was placed a solution of 1-(4-methoxybenzyl)-6-(piperazin-1-yl)-1H-indazole (2 g, 6.21 mmol) in 2,2,2-trifluoroacetic acid (70 mL). To the mixture was added H2O (2.3 mL). The resulting solution was allowed to react, with stirring, overnight while the temperature was maintained at 105° C. in a bath of oil. The reaction progress was monitored by TLC (CH2Cl2/MeOH=5:1). The mixture was concentrated by evaporation under vacuum using a rotary evaporator. The resulting mixture was washed 2 times with 200 mL of ethoxyethane and 2 times with 200 mL of CH2Cl2. This resulted in 1.78 g (crude) of 6-(piperazin-1-yl)-1H-indazole as a grey solid.
