Synthesis of compound 47.5. Into a 100-mL round-bottom flask, were placed compound 47.4 (1 g, 5.88 mmol, 1.00 equiv), dioxane (30 mL) and POCl3 (4.47 g, 29.15 mmol, 5.00 equiv). Reaction was stirred for 1.5 h at 90°C in an oil bath. Upon completion solvent was removed under reduced pressure. The pH value of the solution was adjusted to 8 with saturated sodium carbonate solution. The resulting solution was extracted with 3x100 mL of ethyl acetate, organic layers were combined and dried over anhydrous sodium sulfate and concentrated in vacuum. The crude was purified using flash column chromatography to furnish 550 mg (50%>) of compound 47.5 as a white solid.
