Steps:
2.1 (1) Synthesis of 4-chloro-N-(2-hydroxyphenyl)benzamide (IV-b)
To a 50mL round-bottomed flask was added o-aminophenol II-a (545mg, 5.0mmol), ethyl acetate (20mL) and pyridine (413μL, 5.5mmol) in sequence, at -10°C (-10°C)4-Chlorobenzoyl chloride III-b (636 μL, 5.0 mmol) was added dropwise under conditions. After the dropwise addition was complete, stirring was continued for about 20 minutes, and the reaction was monitored by TLC until the reaction was complete. 10% HCl solution (50 mL) was added to the reaction solution, followed by extraction with ethyl acetate in a separatory funnel (three times, 100 mL each). The organic phase was dried over anhydrous sodium sulfate and concentrated under reduced pressure to obtain a crude product, which was separated and purified by column chromatography (ethyl acetate:petroleum ether=1:4),4-Chloro-N-(2-hydroxyphenyl)benzamide (IV-b) was obtained as a beige solid, 1.15 g, yield: 93%.
