A solution of ethyl imidazo[1,2-a]pyridine-2-carboxylate (11.43 g, 60.1 mmol, J. Org. Chem., 1965, 30, 2403-2407) in glacial acetic acid (100 mL) was treated with W- iodosuccinimide (14.87 g, 66.1 mmol). After stirring at rt for 30 min, the reaction mixture was concentrated to dryness. The residue was taken up in dichloromethane and washed with a 1 :1 :1 mixture of 10% aqueous Na2CO3, brine and saturated Na2S2O3. The aqueous layer was extracted with additional dichloromethane (2x). The combined organic layers were dried over Na2SO4 and concentrated. Flash chromatography (silica gel, 0 to 100% ethyl acetate in hexanes) afforded ethyl 3-iodoimidazo[1 ,2-a]pyridine-2- carboxylate as a pale yellow solid in quantitative yield. 1H NMR (DMSO-c/6): δ 8.41 (d, 1H), 7.62 (m, 1 H), 7.41 (m, 1H), 7.11 (m, 1 H), 4.30 (q, 2H), 1.31 (t, 3H). MS m/z 317 (M+1 ).
