The virus preservation solution is mainly the component added in the virus sampling tube, and not every sample collection place can be detected by PCR, so the collected virus swab samples need to be transported. After the samples are sent to the detection institution, they are analyzed by PCR amplification and other methods, which belongs to the nucleic acid detection of virus.

Detection of VTM virus storage solution

The sampling tube for virus sample collection is composed of sampling tube, sampling swab (nasal swab and pharyngeal swab) and virus preservation solution added in the tube. The preservation solution can be divided into inactivated type and non inactivated type

Inactivated preservation solution: it contains high concentration of lysate, which can not only quickly destroy the cell membrane of virus host cells, but also denature proteins, denaturate and precipitate virus proteins, so that nucleic acids can get rid of protein entanglement and release nucleic acids; it also contains buffer EDTA, Tris and nuclease inhibitors.

Non inactivated preservation solution: Hanks solution is mainly made of various inorganic salts, buffers, glucose and 0.4% phenol red under sterilization conditions, which is consistent with the pH value, osmotic pressure and aseptic conditions of the cell growth state, so as to prolong the survival time of the virus host cells in vitro; in addition, it also contains protein protectants to protect the stability of the virus, as well as buffers and cryoprotectants.

Use of virus preservation solution and sampling tube:

Before sampling, the sample information shall be indicated on the label;

According to different experimental purposes, sampling swabs were used to collect samples at corresponding positions;

The specific sampling methods are as follows:

a) Nasal cavity sampling: insert the swab head into the nasal cavity gently for sampling, use another swab to collect another nasal cavity sample in the same way, immerse the swab head in the sampling liquid and break the tail;

b) Throat collection: throat samples are collected with swabs, and the swab head is also intruded into the sampling solution to break the tail.
