The Chinese name of HEPES: hydroxyethylpiperazine ethanesulfonic acid. The partial formula: C8H18N2O4S. HEPES is a zwitterionic organic chemical buffer, which has no toxic effect on cells. At the same time, it is also a hydrogen ion buffer, which can control a constant pH range for a long time. It has a higher pH value in the range of 7.2-7.4. Good buffering capacity.
There are two ways to use HEPES:
1. Add directly to the configured culture solution according to the required concentration, filter and sterilize, add 2.38 grams of HEPES per 1000 ml of culture solution, adjust the pH to 7.2 with 1N NaOH after dissolution, and use after filtering and sterilizing. At this time, the concentration of HEPES used is 10 mmol/L.
2. Make 100x stock solution (1mol/L), dissolve 23.8g HEPES in 90ml double-distilled water, adjust the pH to 7.5-8.0 with 1N NaOH, then dilute to 100ml with water, filter and sterilize into small bottles, 4 degrees Celsius to Store at 20 degrees Celsius. Before use, take 99ml of culture medium and add 1ml of stock solution to the final application concentration, which is still 10mmol/L.
HEPES is an amphoteric buffer, which is soluble in water and does not form stable complexes with metal ions. It has a good buffering capacity in the pH range of 7.2-7.4. In most cases, it will not interfere with the biochemical process and can be widely used in a variety of biochemical reactions:
1. HEPES is often used as a buffer reagent in the cell culture medium of various types of organisms, cell-cell adhesion, short-term cell aggregation and culture, and buffer for cleaning tissues and cells;
2. In protein research, PIPES is often used as the component of the binding buffer and the elution fluid in cation exchange chromatography;
3. In DNA research, PIPES is used as the buffer solution in the AFM and electroporation experiments of the calcium phosphate and DNA precipitate formation system.
