1、 Introduction to acridine ester NSP series
The acridine ester NSP series is a group of compounds widely used in the fields of biological detection and fluorescence labeling, both of which have high sensitivity and specificity. The following is a brief introduction:
1. Acridine salt NSP-SA: Introducing sulfonamide groups, it has high stability and strong resistance to hydrolysis. It is stable in acidic solutions (pH<4.8) and can be stored at room temperature for 4 weeks with protein conjugates. Its photoquantum yield does not decrease, and the freeze-dried product can be stored for more than one year at -20 ° C. At the same time, changes were made in hydrophilicity by introducing an internal salt of propionic acid, which increased its solubility and made it water-soluble with biomarkers such as antibodies and nucleic acids, enabling luminescence detection to be carried out in water. However, due to its acridine carboxylic acid nature, condensation agents such as EDCI need to be added in order to couple with proteins containing amino groups.
2. Acridine salt NSP-SA-NHS: This is a key fluorescent labeling substance with large steric hindrance, good hydrolysis stability, and high signal-to-noise ratio properties. Its photoquantum yield is superior to acridine ester AE-NHS, and can be applied in direct chemiluminescence analysis and chemiluminescence immunoassay. At the same time, NSP-SA-NHS chemiluminescence does not require any catalyst or enhancer, and can emit light in a dilute alkaline solution with hydrogen peroxide, thereby reducing background luminescence, improving signal-to-noise ratio, and reducing interference factors. In addition, there are many advantages in other places, such as rapid and concentrated light release, fast luminescence efficiency, high luminescence intensity, stable markers, and can be stored for six months at 2-8 ℃.
3. NSP-SA-ADH: It is an internal salt of acridine sulfonamide. Due to the different bond orders between C-N and C-O bonds, the C-N bond is greater than the C-O bond, and acridine amide compounds have strong resistance to hydrolysis. Because it is acridine hydrazide and contains free amino groups, the end of acridine hydrazide is suitable for direct coupling of polysaccharides, nucleic acids, or proteins containing aldehyde groups.
4. Acridine ester NSP-DMAE-NHS: NSP-DMAE-NHS is an ideal immunoassay biomarker for luminescence. The luminescence intensity of the NSP-DMAE-NHS conjugate with antibody (Ab) is higher, and it exhibits flashing luminescence, reaching the ideal peak at 0.4 seconds. Compared to the AE-NHS type, the thermal stability is also significantly improved. For example, at room temperature, in PB buffer with pH 7.0, after two weeks, the luminescence activity only decreased by 3.6%.
