How to prepare N - (2-hydroxyethyl) piperazine N '-3-propanesulfonic acid (EPPS) buffer?
In biochemistry and molecular biology research, buffers are indispensable experimental reagents used to maintain the stability of experimental systems. N - (2-hydroxyethyl) piperazine-N '-3-propanesulfonic acid (EPPS) is a commonly used buffer with an effective pH range of 7.3-8.7 and PKa8.0 (25 ℃). By preparing an EPPS buffer, the environment can be kept consistent within a certain pH range, ensuring the accuracy of experimental results. So, how is it formulated?
Preparation steps of EPPS buffer:
1. Prepare 0.1M EPPS solution (solution A):
(1) Weigh approximately 25.233 grams of EPPS powder and place it in a clean volumetric flask.
(2) Add an appropriate amount of deionized water and set the solution volume to 1000 milliliters (1 liter).
(3) Thoroughly stir until the EPPS powder is completely dissolved.
2. Prepare 0.1M NaOH solution (solution B):
(1) Weigh approximately 4 grams of NaOH powder and place it in a clean volumetric flask.
(2) Add an appropriate amount of deionized water and set the solution volume to 1000 milliliters.
(3) Stir evenly until the NaOH powder is completely dissolved.
3. Mixed EPPS and NaOH solution:
Mix the prepared 0.1M EPPS solution (solution A) and 0.1M NaOH solution (solution B) in the required volume ratio. During the mixing process, it is necessary to stir thoroughly to ensure that the two solutions are thoroughly mixed and uniform.
4. Adjusting pH value:
Add NaOH solution dropwise to the mixed solution, and measure the pH value of the solution using a pH meter to reach the desired range. The pH of EPPS buffer is between 7.3 and 8.7.
Through the above steps, EPPS buffer solution can be successfully prepared and used in various biochemical experiments. Pay attention to using accurate weighing tools during the preparation process to ensure the accuracy of the solution concentration and pH value. At the same time, strictly following the experimental steps and following safe operations can fully ensure the accuracy of the experimental results.
Of course, in addition to considering the details of preparation, the use of EPPS raw materials also needs to be carefully considered. At present, there are many merchants selling EPPS in the market, and to select high-quality EPPS from them, it is necessary to find reliable suppliers. As an established manufacturer of buffering agents, Desheng can provide a large number of stable and inexpensive raw materials with a purity of up to 99%, making it a popular choice for many people.
