The Role of Chemiluminescence Reagents in Immunoassay
Chemiluminescence immunoassay refers to the use of luminescent or enzymatic substances to label antigens and antibodies. After the end of the immune reaction, it emits light through oxidation or enzymatic reactions, and the concentration of the test substance is determined based on the intensity of the light emitted, thereby determining changes in the body. Chemiluminescence immunoassay has attracted much attention both domestically and internationally due to its high sensitivity, wide linear range, long effective time of the marker, and no radioactive hazards.
Different chemiluminescence reagents have different luminescence principles and can be divided into three categories based on the different markers: chemiluminescence immunoassay, chemiluminescence enzyme immunoassay, and electrochemical luminescence immunoassay.
Different immunoassays have different requirements for the methods, instruments, and reagents used. Luminol and acridine ester reagents are commonly used reagents in chemiluminescence immunoassay.
Luminol, Isoluminol, and Luminol Sodium salts were commonly used reagents in the early stages of chemiluminescence immunoassay, mainly used for the determination of small molecules of inorganic and organic matter. Their principle is to catalyze the formation of excited intermediates of 3-aminophthalic acid by horseradish peroxidase under the oxidation of hydrogen peroxide, and emit photons when it returns to the ground state.
Acridine ester reagents can emit light in the presence of hydrogen peroxide and oxides, which has the advantages of simple operation, fast luminescence, no catalysis, and high luminescence efficiency. The luminescence principle is that hydrogen peroxide undergoes an electrophilic addition reaction at the carbon 29 position to generate peroxide, which is decomposed into excited state N2 methylacridone and CO2 through transition state dioxane ketone. The former returns to the ground state and emits about λ= Photons at 430nm.
Luminol and acridine ester reagents have their own advantages, and you can choose according to your own needs when purchasing. Desheng has a history of researching chemiluminescence reagents for nearly 20 years. Luminol, Isoluminol, Luminol Sodium Salt, and Acridine Ester series products are sold with good quality and are highly supported and trusted by domestic and foreign users. Welcome to consult!
