In some analytical experiments, buffer solvent has become an indispensable solution. The reason why it is important is that it has a certain resistance to the interference of external factors due to its special performance. As in the biochemical reaction, a stable acid-base environment is often needed, which cannot be changed due to the addition of external substances, and the buffer solution can achieve the purpose of stabilizing the PH value, so that the test object can survive and react in the appropriate environment, and ensure the experimental process. 1、 Buffer preparation

If it is necessary to prepare a certain acid-base solvent, it is necessary to first select whether it is a weak acidic solution or a strong alkaline solution according to the situation, and its pKa value should also be as close as possible to the pH value of the solution to be prepared, then calculate the acid-base concentration ratio, and then prepare according to this concentration ratio.

 

2、 Buffer preparation

1. PBS buffer

PBS has a wide range of buffer PH, which can be used to prepare solutions with different acidity and alkalinity, and is also a common buffer system. The preparation method of this solution depends on the situation. If it is an acid solution, it can be directly mixed with NaH ? PO ? or KH ? PO ?, and the PH range is 1-5; If it is an alkaline solution, it needs to be prepared with Na  HPO ? or K  HPO ?, with PH range between 9-12.

2. Tris-HCl buffer

If 1M Tris-HCl is used, 1000ml of solution is prepared with PH value ranging from 7.4-8.0, and its preparation method is as follows:

(1) Weigh about 120g of Tris and pour it into a 1000ml beaker;

(2) Then add about 800 ml of purified water and stir it fully until it is completely dissolved;

(3) According to the concentration calculation table, add an appropriate amount of HCl, adjust it to the required PH value, and fix the solution to 1000ml.

(4) Pay attention to storage at room temperature after high-temperature and high-pressure sterilization.

 

3. HEPES buffer

Because its buffer range is within the neutral range and can maintain a stable PH value when the container is open, it is often used in cell culture and biochemical diagnostic reagents. If 1M solution with pH 7.0 is to be prepared, the preparation method is as follows:

(1) Weigh about 120g of HEPES powder and pour it into the container;

(2) Use purified water to fully dissolve it in a 400mL bottle;

(3) Add NaOH solution for mixing, and then adjust the pH value to 7.0;

(4) Keep the solution at a constant volume of 500 mL and store it at room temperature. Pay attention to the preparation and use immediately;
