PIPES is a member of the ethanesulfonic acid buffer series. The Chinese name is 1-4-piperazine diethanesulfonic acid. It is one of the ethanesulfonic acid series buffer reagents developed by Good et al. in 1960. It has some of Good's buffers. Important features such as medium pKa, maximum water solubility and minimum solubility in all other solvents, minimum salt effect, minimum pKa change with temperature, (PIPES is hardly soluble in water) chemically and enzymatically stable, minimum absorption of visible light or ultraviolet spectra Range, easy to synthesize and other features.
The effective buffer range of PIPES is pH 6.1-7.5, pKa=6.8 (25°C). Its pKa is close to physiological pH, suitable for cell culture. In addition, in animal and plant chemistry, PIPES can also be used as a buffer reagent to minimize lipid damage and experimental artifacts caused by glutaraldehyde fixation. It is worth noting that because PIPES can form free radicals, it is not suitable for oxidized reducing substances.
PIPES can be used in in vitro cell culture media. In the study of the determination of lactic acid in plasma by the lactate oxidase method, it was found that PIPES can be stable for 172 hours at 37°C. For immunofluorescence labeling, 4% paraformaldehyde prepared in 50mmol/L PIPES buffer can be used for immobilization. PIPES can meet the requirement of extremely low complexing of divalent cations when adenosine diphosphate (ADP) is bound to the relaxed fan-forming muscle fiber cells.
