Synthesis of Centhaquine Citrate

Centhaquine Citrate is synthesised by the reaction of Styrenyl quinoline and d piperazine. The specific synthesis method is as follows:

Styrenyl quinoline and piperazine were combined in ethanol and acetic acid and heated at reflux. After an aqueous workup, centhaquine was isolated by recrystallization from hexane and then converted to its citrate salt with an overall yield of 66% for the two-step sequence.

Centhaquine citrate is an α2B adrenergic receptor agonist developed by Pharmazz Inc. and was approved by the Central Drugs Standard Control Organization of India for the treatment of hypovolemic shock due to blood loss. The drug is administered at a dose of 0.01 mg/ kg as an intravenous infusion in normal saline. In patients experiencing hypovolemic shock, centhaquine decreases blood lactate, increases blood pressure, increases cardiac output, and improves tissue perfusion. Centhaquine citrate also restores renal blood flow and protects against tissue damage after hemorrhagic shock.
