Does nanodispersion-based paclitaxel injection have better antitumor effects than paclitaxel?
Dec 16,2024
Yes. Currently, researchers have developed a nanoparticle-based concentrate for injection (Nab-paclitaxel), which is a water-soluble formulation of paclitaxel. Paclitaxel binds to tubulin and inhibits the disassembly of microtubules, thereby inhibiting cell division. Compared with paclitaxel alone, the nanodispersion-based formulation uses less toxic solvents and a larger dose, resulting in higher paclitaxel concentrations at the tumor site and better safety.
In addition, they compared the antitumor efficacy and toxicity of Nab-paclitaxel with paclitaxel in vitro and in a mouse endometrial cancer model. Nab-paclitaxel is more effective than paclitaxel in inhibiting cancer cell growth. Nab-paclitaxel also causes apoptosis and cell cycle G1 arrest more effectively than paclitaxel. Similar effects were observed in a mouse model of endometrial cancer. However, it remains to be verified in clinical applications.

Paclitaxel is a chemotherapeutic agent marketed under the brand name Taxol among others. Used as a treatment for various cancers, paclitaxel is a mitotic inhibitor that was first isolated in 1971 from the bark of the Pacific yew tree which contains endophytic fungi that synthesize paclitaxel. It is available as an intravenous solution for injection and the newer formulation contains albumin-bound paclitaxel marketed under the brand name Abraxane.
