產(chǎn)品用途
Afoxolaner用于治療和控制狗的跳蚤和蜱蟲(chóng)感染。在被狗攝入后，阿伏奧利內(nèi)會(huì)分布在狗的全身。當(dāng)跳蚤或蜱蟲(chóng)叮咬狗時(shí)，它們會(huì)接觸到這種藥物，并在吸食血液時(shí)被殺死。
它作為一種拮抗劑作用于配體門控氯離子通道，特別是那些由神經(jīng)遞質(zhì)γ -氨基丁酸（gaba受體）門控的通道。在氯離子通道調(diào)節(jié)劑中，異惡唑啉類與昆蟲(chóng)gaba門控氯離子通道內(nèi)一個(gè)獨(dú)特的靶點(diǎn)結(jié)合，從而阻斷氯離子跨細(xì)胞膜的突觸前和突觸后轉(zhuǎn)移。長(zhǎng)時(shí)間的阿伏唑啉引起的過(guò)度興奮導(dǎo)致中樞神經(jīng)系統(tǒng)活動(dòng)失控和昆蟲(chóng)和螨蟲(chóng)死亡。

Applications
Afoxolaner is used to treat and control flea and tick infestations in dogs. After being ingested by a dog, afoxolaner is distributed throughout the dog’s body. When fleas or ticks bite the dog, they are exposed to the drug and killed during their blood meal.
It acts as an antagonist at ligand-gated chloride channels, in particular those gated by the neurotransmitter gamma-aminobutyric acid (GABA-receptors). Isoxazolines, among the chloride channel modulators, bind to a distinct and unique target site within the insect GABA-gated chloride channels, thereby blocking the pre- and post-synaptic transfer of chloride ions across cell membranes. Prolonged afoxolaner-induced hyperexcitation results in uncontrolled activity of the central nervous system and death of insects and acarines.
