背景（Background）
谷氨酸脫羧酶（GAD）是一種催化谷氨酸脫羧為γ氨基丁酸（GABA）的酶，γ氨基丁酸是高級腦區(qū)的主要抑制性遞質，也是胰島中推測的旁分泌激素。GAD使用磷酸吡哆醛（PLP）作為輔因子。GAD的兩種分子形式（65 kDa和67 kDa）高度保守，均在中樞神經(jīng)系統(tǒng)、胰島細胞、睪丸、輸卵管和卵巢中表達。這些異構體在大鼠和小鼠的大腦中呈區(qū)域性細胞質分布。GAD65是一種ampiphilic，膜錨定蛋白（585 a.a.），負責囊泡GABA的產(chǎn)生。GAD67是細胞質（594 a.a.），似乎負責大量的細胞質GABA產(chǎn)生。
