背景（Background）

信號轉(zhuǎn)導(dǎo)子和轉(zhuǎn)錄激活子3（STAT3）調(diào)節(jié)對基本細(xì)胞過程（包括存活、增殖、自我更新、血管生成和免疫反應(yīng)）重要的基因的表達(dá)。作為早期腫瘤診斷標(biāo)志物，組成型STAT3激活被認(rèn)為是模型系統(tǒng)中引起惡性轉(zhuǎn)化和腫瘤發(fā)展的充分因素。癌癥細(xì)胞中經(jīng)典的IL6/STAT3信號通路。IL-6與膜結(jié)合的IL-6受體α（IL-6R）和IL-6受體β（也稱為gp130）結(jié)合。IL-6/IL-6R/gp130復(fù)合物激活JAKs的磷酸化，隨后是STAT3的磷酸化和激活。然后，磷酸化的STAT3形成同二聚體并易位到細(xì)胞核中，與靶基因的啟動子區(qū)域結(jié)合并激活靶基因轉(zhuǎn)錄。
