MCA-AVLQSGFR Lys (Dnp) Lys NH2 is a FRET substrate peptide of the main protease of severe acute respiratory syndrome coronavirus (SARS-CoV Mpro), with a sequence containing 10 amino acids. The substrate sequence of MCA-AVLQSGFR-Lys (Dnp) - Lys-NH2 is derived from the residue P4-P5 'of SARS-CoV Mpro N-terminal automatic processing site with AVLQSGFRK sequence. The enzymatic cleavage between Q4 and S5 of MCA-AVLQSGFR-Lys (Dnp) - Lys-NH2 explains that it emits a highly fluorescent 7-neneneba methoxy Coumarin (Mca) group, which is affected by the internal quenching agent of 2,4-dinitrophenyl (Dnp), resulting in a large increase in fluorescence intensity.
