90% To a solution of <strong>[1190309-71-7]3-bromo-5-fluoro-1H-pyrrolo[2,3-b]pyridine</strong> (0.74 g, 2.0 mmol) in THF (5 mL) was added NaH (127 mg, 3.0 mmol) at 0 C, and the mixture was stirred for 30 mm maintaining at this temperature, then TsC1 (458 mg, 2.4 mmol) was added. The resulting mixture was warmed to it and stirred overnight. The reaction mixture was quenched with water (50 mL), and the resulting mixture was partitioned. The aqueous layer was extracted with ethyl acetate (50 mL x 2). The combined organic layers were washed with saturated brine (80 mL), dried over anhydrous sodium sulfate, and filtered. The filtrate was concentrated in vacuo, and the residue was purified by silica gel column chromatography (PE/EtOAc (v/v) = 4/1) to give the title compound as a yellow solid (740 mg, 90%).MS (ESI, pos. ion) m/z: 370.8 [M+H]1H NMR (400 MHz, CDC13) (ppm): 8.32 (s, 1H), 8.06 (d, J = 8.3 Hz, 2H), 7.84 (s, 1H), 7.47 (dd, J = 7.8, 2.6 Hz, 1H), 7.29 (d, J = 8.3 Hz, 2H), 2.38 (s, 3H).

After 1h, a solution of p-TsCl (5.7 g, 30 mmol) in DMF (20 mL) was added slowly at the same temperature and stirred for 2 h. After completion of the reaction as indicated by TLC, the mixture was poured into ice cold water (200 mL) and filtered the precipitated solid and dried to afford 3-bromo-5-fluoro-1-tosyl-1H-pyrrolo[2,3-b]pyridine (6) (6.2 g, 16 mmol, 76 %) as an off-white solid.
