PZII-029 Chemical Properties,Uses,Production
Uses
PZ-II-029 is a GABAA positive allosteric modulator that selectively binds with high affinity to α6β3γ2. PZ-II-029 shows anti-migraine effects[1].
Biological Activity
PZ-II-029 is the first compound to be identified as a specific postitive modulator of GABAA channels composed of α6β3γ2 subunits.
in vivo
PZ-II-029 (1-10 mg/kg; i.p.; 30 min before Capsaicin instillation) inhibits central and peripheral trigeminal responses in a rat migraine model[1].
| Animal Model: | Male Wistar rats (250-300 g; 8-10 weeks) induced by intra-cisternal (i.c.) instillation of Capsaicin[1]. |
| Dosage: | 1, 3, or 10 mg/kg |
| Administration: | i.p.; 30 min before Capsaicin instillation |
| Result: | Significantly attenuated the trigeminal cervical complex (TCC) neuronal activation and trigeminal ganglia (TG) calcitonin gene-related peptide immunoreactivity (CGRP-ir) elevation, and dural CGRP depletion induced by capsaicin. |
