纖維蛋白肽A(人)是在凝血酶作用下,纖維蛋白原α(A)鏈的精-16和甘-17之間的肽鏈裂解,釋放出由1~16個(gè)氨基酸組成的FPA。是反映體內(nèi)凝血活性及纖維蛋白最終形成血栓的可靠指標(biāo)。血纖維蛋白肽A(人)為在纖維蛋白原活化過(guò)程中,最先由纖維蛋白原α鏈上水解下的片段。
血纖維蛋白肽A(人)的前體蛋白為Fibrinogen。Fibrinogen中含有一信號(hào)肽,由19個(gè)氨基酸組成,位于原蛋白的1~19區(qū)域。因?yàn)楹行盘?hào)肽的蛋白質(zhì)一般能夠被分泌到胞外,作為細(xì)胞因子起作用,所以可以認(rèn)為Fibrinogen是一個(gè)分泌蛋白。原蛋白被分泌到胞外時(shí),信號(hào)肽被剪切留在胞內(nèi),剩下的位于20~664區(qū)域的氨基酸鏈則被分泌到胞外成為兩個(gè)成熟蛋白,分別為20~35的血纖維蛋白肽A(人)和35~664的Fibrinogen,alphachain,isoformalpha。血纖維蛋白肽A(人)的氨基酸數(shù)目為16個(gè),相對(duì)分子質(zhì)量為1536.5,等電點(diǎn)為3.92;負(fù)電荷殘基總數(shù)(Asp+Glu)為4;正電荷殘基總數(shù)(Arg+Lys)為1;半衰期為4.4h;不穩(wěn)定系數(shù)為22.11,該蛋白分類(lèi)為穩(wěn)定蛋白;脂肪系數(shù)為55.00;親水性評(píng)估為-0.431。
The conversion of monomeric fibrinogen into polymeric fibrin is mediated by thrombin, which binds to the central region of fibrinogen and catalyzes cleavage of the 2 short peptides, the 16-residue fibrinopeptide A (FpA) and the 14-residue fibrinopeptide B (FpB), located at the NH 2 -termini of the Aα and Bβ chains, respectively.
Fibrinopeptide A (FPA) is a small polypeptide cleaved from fibrinogen by thrombin, has a short half-life, and is considered a sensitive biochemical marker of thrombin activity, fibrin generation, and ongoing thrombosis.
