Dapansutrile is an orally active β-sulfonyl nitrile compound that selectively inhibits the NLRP3 inflammasome.Upon oral administration, dapansutrile specifically binds to and inhibits NLRP3, the sensor component of the NLRP3 inflammasome, and prevents the formation of the NLRP3 inflammasome. This inhibits the production of the pro-inflammatory cytokines interleukin-1-beta (IL-1beta) and interleukin-18 (IL-18), which may prevent or reduce inflammation. Dapansutrile is a NLRP3 inflammasome inhibitor that disrupts inflammasome formation. Reduces LPS/ATP-induced IL-1β secretion in human and mouse macrophages in vitro, without affecting TNFα levels. Reduces LPS-induced inflammation in vivo. Also reduces joint swelling and markers of inflammation in synovial fluid in a mouse model of acute arthritis. Displays cardioprotective effects in a mouse model of cardiac ischemic reperfusion injury. | [Side effects]
