Thiazoloquin(az)olin(on)es, such as the small molecule 78c, have been identified as potent CD38 inhibitors (CD38i). CD38 inhibitor 78c is a potent, reversible, and uncompetitive inhibitor of CD38. 78c reverses age-related NAD+ decline and ameliorates several metabolic, structural, and molecular ageing features in chronologically aged and progeroid mice. The CD38 inhibitor 78c increases naturally aged mice's lifespan and health span. In addition to a 10% increase in median survival, 78c improved exercise performance, endurance, and metabolic function in mice. The effects of 78c were different between the sexes[1-2].
